Run batch tomograms

Goal:

Acquire all selected and setup batch positions with a standard tilt scheme.

Checks

Enough nitrogen in the tank?
Autoloader turbo off?
Enough disk space on Gatan PC X: drive?
Objective aperture in and centered?
Astigmatism ok?
Dose rate ok?
Frame saving on?
Column valves open?
VPP: Phase plate inserted?
VPP: Aperture control panel vissible and flapped out in UI?

The Krios uses 25 to 30 liters of liquid nitrogen per day, the big tank is about 200 liters

At the level shown here, just replace the tank.

Focus range

Focus range is handled by two scripts, one run before each tilt series and one during each tilt.

Edit the focus range in script PreTomo:

Check in Tilt Series menu if Run Script in TS is selected:

Run

SerialEM file menu, deselect continuous saving for the log file, close the log file. Each tilt series will get its own log file.

Navigator menu, Acquire at points.
Run script PreTomo.
Select Primary task Acquire tilt series.
Run script after PostTomo
Select Close column valves at end.
Select Send email at end.

GO!

You will receive an email per tilt series, either succesfull or error, error can however also mean the full tilt range was not reached. If there is an error email about another problem, or is the emails stop coming in, you ned to start worrying!
