Goal:
- Load samples.
- Maximum is 12.
- To properly screen, 4 to 6 will be enough to handle in a day.
Load grids into cartridges
Turn on the Leica heater for the tools.
Fill in the loading chamber with liquid N2, do not overpass the middle part.
Put the cartridge on the loading chamber; rotate the upper part of the
loading chamber using a large screwdriver. Insert another cartridge and rotate
again.
Once you have inserted all the cartridges cover the chamber with its lead.
Put on the protection mask in order to do not contaminate your sample (your breath is warm and full of water…).
In the loading chamber put the circular box where you will put the loaded cartridges.
Take a clip ring and insert it in the apposite tool. Put the tip of the
tool on the table and press down the back part once (the clip ring
should be now in the correct position in the tip).
Cool down the tweezers, take the grid and put it on the cartridge.
Rotate the loading chamber lead counterclockwise.
Cool down the tool with the clip ring.
To insert the clip ring push the back of the clip ring tool.
Rotate the loading chamber lead counterclockwise and load the other grids.
Load cartridges into cassette
Fill with liquid N2 the transfer box. The liquid N2 should not go into the chambers for circular grid boxes.
Rotate the loading chamber lead clockwise.
Using the apposite forceps take the loaded cartridges (cartridge+grid+clip ring) and put them into the circular grid box.
Transfer the grid box into the transfer box.
Transfer the loaded cartridges into the cassette. It is better if you leave an empty space between each other.
Important: be sure they are completely inserted, slightly push them on the top.
Load cassette into capsule
Fill the capsule with liquid N2.
Remove the bottom lead of the transfer box then insert the capsule.
Pull the small metal pin on top of the capsule to be sure that the cassette is in place.
Temperature control status
Check that the Krios temperatures are all in the green range:
Dock cassette to autoloader
Insert the capsule into the Krios and push the green button. Take the
capsule back once the loaded cartridge had been taken from the Krios.

How to leave the transfer equipment
Put all the transfer equipment (loading chamber,
transfer box and capsule) in the heating cabinet.
DO NOT CHANGE HEATING CABINET TEMPERATURE!!!
