Mapping Grids
Goal:
- Setup microscope.
- Start SerialEM macro to automatically:
- Wait some time for the loader to cool down again.
- Do AutoLoader Inventory.
- Map all grids found by the inventory at lowest magnification.
Titan UI:
- Column valves must be closed!

- Condenser 2 aperture 50 micrometer should be in.
Objective aperture must be out!
SerialEM:
- Set your email address, menu Tilt Series/Set Email Address.
- Navigator, Open imaging States.
- Load imaging state, double click on "Grid map" (EFTEM LM 135, spot 6).

- Check in Titan UI if the setting is ok, mag should me LM 135!!!


- Open column valves in SerialEM:
- Titan FluCam viewer, Insert Screen, check if the beam is centered around the GIF entrance aperture (small green circle).
- Insert the beam stop halfway in with
, things should now look like this (the right edge is not a problem):

- Take a record image in SerialEM, image should look like this:

- FluCam viewer, retract the beam stop with
.
- SerialEM Navigator menu, Open.
- SerialEM Navigator menu, Montaging & Grids, Setup Full Montage .

Do not select "Skip this dialog in future (re-enable in file menu)"!
- Macro menu, Edit 1, change the delay time if needed (depends on autoloader temperature, Krios 2 +/- 10 min).
- Close column valves.
- Set your email address in menu Tilt Series/Set email address.
- Run Macro 1, takes a while.
- Save Navigator file, Navigator menu, Save.
- Choose the grid you want to image, select the grid in the Titan
UI AutoLoader control panel, click load (any loaded grid will
automatically be unloaded).
